I would like to know why does the ROI not adjust to a new ROI after being defined in Mech Eye.

It is because of the Capture Mode you used.
For Mech-Eye 2.1.0, the ROI is only effective when the Camera 1 mode is selected, and is ineffective when the Merge mode is selected.
